Purified from lneutrophilic polymorphonuclear leukocytes, white blood cells found in peripheral blood, and lyophilized. The protein is greater than 96 percent pure by SDS-PAGE and is free of cathepsin G, MPO and lysozyme. One unit is defined as the amount of enzymatic activity which releases 1 micromol of p-nitroanilide per minute from MeO-Suc-Ala-Ala-Pro-Val-pNa at 25C, pH 8.0.

The neutrophil form of elastase is 218 amino acids long, with two asparagine-linked carbohydrate chains. Current studies show that both Cathepsin G and Neutrophil Elastase are key enzymes for tissue injury caused by activated neutrophils, such as occurs in Acute Lung injury.
